- ABCC Subfamily Vacuolar Transporters are Involved in Pb (Lead) Detoxification in Saccharomyces cerevisiaePublication . Sousa, Cátia A.; Hanselaer, Simon; Soares, Eduardo V.The present work has as objective to contribute for the elucidation of the mechanism associated with Pb detoxification, using the yeast Saccharomyces cerevisiae as a model organism. The deletion of GTT1 or GTT2 genes, coding for functional glutathione transferases (GST) enzymes in S. cerevisiae, caused an increased susceptibility to high Pb concentrations (500-1000 μmol L(-1)). These results suggest that the formation of glutathione-Pb conjugate (GS-Pb), dependent of GSTs, is important in Pb detoxification. The involvement of ATP-binding cassette (ABC) vacuolar transporters, belonging to class C subfamily (ABCC) in vacuolar compartmentalization of Pb, was evaluated. For this purpose, mutant strains disrupted in YCF1, VMR1, YBT1 or BPT 1 genes were used. All mutants tested, without vacuolar ABCC transporters, presented an increased sensitivity to 500-1000 μmol L(-1) Pb comparative to wild-type strain. Taken together, the obtained results suggest that Pb detoxification, by vacuolar compartmentalization, can occur as a result of the concerted action of GSTs and vacuolar ABCC transporters. Pb is conjugated with glutathione, catalysed by glutathione transferases and followed to the transport of GS-Pb conjugate to the vacuole by ABCC transporters.

This trend is mainly explained by the fact that there is an increasingly active and ageing population in developed countries. The growing demand for this area will consequently lead to the need for many resources, both human and economic. Currently, there are several bionic hands with various uses and functions, and most of these hands are programmed to respond to the movements of a particular user. Besides, most of the applications of medical robotics are mainly directed to the area of surgery. With this proposal, we intend to simplify the whole physiotherapy or rehabilitation process. For that, we have developed an equipment capable of helping the patient to perform the exercises in the comfort of his home, but in an equally efficient way. The operation of the system consists of autonomous use by the patient, who must wear the glove and replicate the exercises, predefined for each user, performed by the biomechanical hand. Having the possibility to define the exercises, ABHA is transferable and can be used by more than one patient.
- Acid pretreatment of sugarcane biomass to obtain hemicellulosic hydrolisate rich in fermentable sugarPublication . Morais Júnior, Wilson Galvão; Pacheco, Thályta F.; Corrêa, Priscila S.; Martins, António A.; Mata, Teresa M.; Caetano, NídiaThe objective of this work was to find the ideal pretreatment conditions with high efficiency to obtain a hydrolyzate rich in fermentable sugars and low possible inhibitors levels. Thus, it was applied diluted phosphoric acid to pretreat the sugarcane biomass. Through a Central Composite Design, it was evaluated the influence of temperature, operating time and acid concentration. The pretreatment efficiency was verified by the concentration of total monosaccharides in the liquid fraction after the reaction. The phosphoric acid concentration of 4.95% at 80 °C, during 375 min, resulted in a hemicellulosic hydrolyzate with the highest concentration of fermentable sugars (saccharification greater than 99%), with the absence of HMF and furfural, and relatively low amounts of acetic acid.

Este artigo descreve vários problemas encontrados em aulas laboratoriais, em cursos de ensino superior, na utilização de laboratórios remotos baseados em sistemas PXI, utilizando o sistema VISIR ou uma solução alternativa. São identificados e explicados três problemas que foram relatados por professores que deram apoio aos estudantes na utilização dos laboratórios remotos. O primeiro problema prende-se com a necessidade de permitir aos alunos selecionar a localização específica onde um amperímetro deve ser inserido nos circuitos elétricos, mesmo que incorreta, replicando as dificuldades do mundo real. O segundo é causado por falhas de sincronismo quando são necessárias várias medições em intervalos curtos, como no ciclo de descarga de um condensador. E o último problema é provocado pelo uso de um multímetro em modo DC na leitura de grandezas em CA, um procedimento que colide com as definições do equipamento. Todos os cenários são apresentados e discutidos, incluindo a solução encontrada para cada caso. A conclusão que se retira do trabalho descrito é que a área de laboratórios remotos é um campo em expansão, onde a sua utilização prática permite o aperfeiçoamento e a evolução das soluções disponíveis, exigindo uma cooperação e partilha de informação entre todos os intervenientes, i.e. investigadores, professores e alunos.
- Adapting remote labs to learning scenarios: case studies using VISIR and RemotElectLabPublication . Fidalgo, André Vaz; Alves, Gustavo R.; Marques, Maria A.; Viegas, C.; Costa-Lobo, Cristina; Henandez-Jayo, Unai; Garcia-Zúbia, Javier; Gustavsson, IngvarRemote laboratories are an emergent technological and pedagogical tool at all education levels, and their widespread use is an important part of their own improvement and evolution. This paper describes several issues encountered on laboratorial classes, on higher education courses, when using remote laboratories based on PXI systems, either using the VISIR system or an alternate in-house solution. Three main issues are presented and explained, all reported by teachers, that gave support to students' use of remote laboratories. The first issue deals with the need to allow students to select the actual place where an ammeter is to be inserted on electric circuits, even incorrectly, therefore emulating real-world difficulties. The second one deals with problems with timing when several measurements are required at short intervals, as in the discharge cycle of a capacitor. In addition, the last issue deals with the use of a multimeter in dc mode when reading ac values, a use that collides with the lab settings. All scenarios are presented and discussed, including the solution found for each case. The conclusion derived from the described work is that the remote laboratories area is an expanding field, where practical use leads to improvement and evolution of the available solutions, requiring a strict cooperation and information-sharing between all actors, i.e., developers, teachers, and students.
- Adding value [valorization] to tannery fleshings: part II – effect of protein hydrolysates on leather dyeingPublication . Bragança, Idalina; Nouws, Henri P. A.; Crispim, AlfredoThe tanning industry generates a high quantity of solid wastes. Therefore, there is a need to create valorization [added value] options for these wastes. The main objective of the present work was to study the effect of protein hydrolysates (HP) prepared from fleshings on leather dyeing. During previous studies it was found that the application of HP products, obtained from fleshings, in leather retannage intensified the colour of crust leather. In this work the CIELAB colour system was used to evaluate the effect of HP on retannage processes. The main conclusions of this study were: (i) HP can be used instead of a dicyanodiamide resin (Fortan DC) if the colour parameters of the standard procedure are to be maintained, and (ii) the replacement of an acrylic resin (Fortan A40) by glutaraldehyde-modified HP (GHP) results in a darker skin, and can therefore be interesting for the reduction of the quantity of dye used.
- Adding Value to Tannery Fleshings Part I – Oils and Protein Hydrolysates – Production and ApplicationPublication . Therefore, there is a need to create valorization [added value] options for these wastes. The present work had as its main objective creating added value by production of fat and hydrolyzed protein. To this end, fleshings were treated by hydrolysis with regard to the influence of various factors. The best result was found for a temperature of 60°C, 4 hours of hydrolysis, 2% of enzyme and 100% of water, with a fat extraction yield of 93%. The fat obtained through the hydrolysis process was used to produce sulphated oils. The protein hydrolysate was concentrated to about 40% of solids content and used to prepare co-products of protein hydrolysate and glutaraldehyde. Sulphated oils were applied in leather fatliquoring and the different protein hydrolysates were tested in leather retannage. The leather samples obtained were evaluated by physical-mechanical tests and the results were compared to those obtained through a standard process. The results were very satisfactory and, in some cases better than the specified standard. The process of adding value to this waste results in a double advantage for the leather industry, reducing the environmental impact and allowing production of alternative products for leather fatliquoring and retannage.
- Addressing software impact in the design of remote laboratoriesPublication . Garcia-Zúbia, Javier; Orduña, Pablo; Lopez-de-Ipina, D.; Alves, Gustavo R.Remote Laboratories or WebLabs constitute a first-order didactic resource in engineering faculties. However, in many cases, they lack a proper software design, both in the client and server side, which degrades their quality and academic usefulness. This paper presents the main characteristics of a Remote Laboratory, analyzes the software technologies to implement the client and server sides in a WebLab, and correlates these technologies with the characteristics to facilitate the selection of a technology to implement a WebLab. The results obtained suggest the adoption of a Service Oriented Laboratory Architecture-based approach for the design of future Remote Laboratories so that client-agnostic Remote Laboratories and Remote Laboratory composition are enabled. The experience with the real Remote Laboratory, WebLab-Deusto, is also presented.
- Adjusting Higher Education Competences to Companies Professional NeedsPublication . Alves, Joaquim; Lima, Natércia; Alves, Gustavo R.; García-Peñalvo, Francisco J.This work reports the incorporation result of a Work Integrated Learning (WIL) Program in the two- years master’s degree in Instrumentation and Metrology Engineering (MEIM) held at the School of Engineering, Polytechnic of Porto (ISEP). The procedure developed to find internships in industrial companies and institutions, which have needs for engineers in the areas of instrumentation and metrology is displayed. The authors also present the information on the situation of students that, by the end of the first academic year, could get enrolled in the curricular unit Dissertation/Project/ Professional Training (opting either for a professional internship in a company or by a research project conducted in ISEP laboratories). In order to understand the benefits and impact of a WIL program, from a student perspective, students were asked to answer a questionnaire. Lastly, the results achieved during the study period are presented, including the student reported outcomes (from the questionnaire), as well as the informal feedback from students and companies’ supervisors. The good results achieved, regarding the development of theoretical-practical competencies as well as generic or soft skills, during the master’s, were also confirmed by the fact that students are hired by the companies to continue their work after the end of professional training.
- Adopting building automation in weblabsPublication . Costa, Ricardo J.; Alves, Gustavo R.; Santos, Domingos S.Several companies have been developing domotic Stds. for building automation, enabling users to locally and remotely control several home devices, like: lights, power sockets, heating, ventilation, and air conditioning systems, among others. Besides contributing to improve the building comfort, these Stds. may also be adopted for other purposes, namely in weblabs used in sciences and engineering remote experiments. To increase the sense of immersion in weblabs, we identify domotic Stds. as a standard solution for turning on/off the power infrastructure and controlling the light and temperature conditions of the physical space where a specific experiment may run, thus approaching the sense of being in the lab facilities while accessing them through the corresponding weblab interface. After identifying the added value to weblabs in terms of power savings and in the control of the environmental conditions, we used our knowledge and the WWW to conduct an extensive search on domotic Stds., and after analysing the results obtained, we choose the most appropriated one to be implemented in a Weblab. Regarding the adopted Std., a proof-of-concept is also described, enabling the control of an halogen lamp and a power socket, using a specific Web interface.
