- Alteração do perfil lipídico em pacientes com esquizofreniaPublication . A terapêutica farmacológica evoluiu nas últimas décadas, sendo prescritos atualmente a estes pacientes antipsicóticos que têm permitido a sua melhor integração na comunidade. No entanto, os antipsicóticos apresentam efeitos colaterais, como alterações no metabolismo da glicose e dos lípidos que, por sua vez, aumentam o risco de síndrome metabólica (SM) e de doenças cardiovasculares nestes pacientes.

Mota, Sílvia; Lopes, Maria; Lamas, Maria Céu; Sousa, MariaA vitamina D é uma hormona que atua diretamente na saúde óssea, no crescimento celular saudável, na imunidade, na musculatura, no metabolismo e noutros sistemas, como o sistema cardiovascular e o sistema nervoso. A sua formação implica uma primeira hidroxilação no fígado, pela 25-hidroxilase, a 25-OHD. Assim, o nível sérico de 25-OHD é o melhor indicador do conteúdo corporal de vitamina D. Um número crescente de estudos tem vindo a associar a insuficiência de vitamina D com aumento do risco para diversas patologias, como doenças cardiovasculares.

É o marcador ecográfico de maior sensibilidade no rastreio de anomalias cromossómicas, sendo associado o seu aumento a achados citogenéticos como Trissomia 21,18 e 13, Monossomia do X e triploidias.
- Antimicrobial susceptibility of Staphylococcus aureus and coagulase-negative staphylococci in oral and nasal cavitiesPublication . Sousa, Marta; Sampaio Maia, Benedita; Costa, Carolina; Campos, Carla; Campos, JoanaStaphylococcus infections are becoming increasingly difficult to treat due to the growing number of resistant strains to old and new antibiotics. Staphylococcus aureus is referenced as the most clinically relevant but other species, known as coagulase-negative staphylococci (CoNS), have emerged as major causes of serious infections. Although nostrils are considered Staphylococcus preferred host habitat, the oral cavity has also been appointed as a relevant reservoir. Thus, this study aimed to evaluate the antibiotic susceptibility (AS) of nasal and oral staphylococci isolates. Therefore, AS was determined to 102 nasal and 90 oral isolates, previously recovered from dentistry students, by disc diffusion agar for amoxicillin, cefoxitin, ciprofloxacin, chloramphenicol, clindamycin, erythromycin, gentamicin, quinupristin-dalfopristin, tetracycline, and trimethoprim+sulfamethoxazole following the EUCAST/CLSI guidelines. In either nasal (N) or oral (O) cavities, the Staphylococcus isolates included 22% of S. aureus and 78% of CoNS isolates, namely S. epidermidis (41%-N,37%-O), S. warneri (15%-N,16%-O), S. capitis (10%-N,4%-O), S. saprophyticus (9%-N,6%-O), S. pasteuri (7%-O), S. lugdunensis (3%-O), and other (S. haemolyticus, S. hominis, S. sciuri, S. cohnii, and S. condimenti/S. carnosus). All S. aureus isolates presented resistance to at least one antibiotic, and 36% in oral and 25% in nasal cavities presented multidrug resistance (MDR). Among CoNS 65% in oral and 49% in nasal cavities presented resistance to at least one antibiotic, and ~5% presented MDR, namely isolates of S. epidermidis, S. pasteuri, S. haemolyticus, and S. hominis. In conclusion, this study revealed the relevance of the oral cavity as a colonization site for Staphylococcus, including S. aureus, and as a potential source/reservoir of antibiotic resistance, which has been widely neglected so far, highlighting the need for oral cavity inclusion in the Staphylococcus carriage and antibiotic screening.

- Caracterização do perfil hematológico de uma população envelhecida em função da sua atividade física e estado nutricionalPublication . O processo de envelhecimento implica alterações ao nível da constituição corporal, sendo frequentes alterações no estado nutricional, na capacidade física e motora, refletidas na maior incidência e progressão de patologias nesta população. Assim, é importante identificar fatores modificáveis que permitam melhorar ou manter a qualidade de vida desta população. O estado de saúde do indivíduo pode ser avaliado através de indicadores analíticos, incluindo o hemograma.
- Decoding immunomodulatory interactions: Exploring the crosstalk between bone marrow-derived mesenchymal stem cells (BM-MSCs) out the bag and peripheral blood mononuclear cells (PBMCs) for potential in-vivo immunosuppressionPublication . Monteiro, Raquel L.; Lopes, Sérgio M.; Roncon, SusanaAllogeneic hematopoietic stem cell transplant is vital for haematological diseases, yet chronic complications arise as donor immune cells start attacking patient tissues. A lab barrier to studying this alloreactivity is the absence of the hematopoietic stem cell niche. Here, we employed BM-MSCs from discarded collection bags to solve this hurdle. The aim was to modulate PMBC proliferation in vitro, downregulating alloreactivity and proposing immunomodulation mechanisms. Responder PBMCs were labelled with CFSE dye, and stimulator cells irradiated. Co-cultured in a 2:1 ratio to mimic alloreactivity in MLR assays (n=3) for 6 days, with or without BM-MCs. MSC phenotype examined pre- and post-co-culture. Flow cytometry examined PBMC proliferation through CFSE histograms and assessed surface marker expression of CD45+, CD3+, CD4+, CD8+, CD19+, CD56+ and CD14+. Resazurin assays tracked metabolic activity. CFSE immunofluorescence recorded PBMC proliferation, while phase-contrast images showed PBMC-MSC physical interactions. Data is shown as mean ± standard deviation. Statistical significance assessed with Two-Way ANOVA test (p<0.05). BM-MSCs expanded successfully, maintaining ideal phenotype and function. CD44+ expression increased significantly in MSCs post-culture. CFSE expression aligned only with PBMC format, whilst phase-contrast images revealed closer alignment in co-culture. Metabolic activity showed enhanced metabolism in PBMCs/MSCs co-culture, with a 15% reduction in proliferating responder cells. March's pending immunophenotyping results will enhance understanding of the observed immunosuppression. Expanding BM-MSCs from discarded bags unravelled immunosuppression in alloreactive conditions. BM-MSCs, lacking CD44, had increased expression due to in-vitro cultivation. Reduced alloreactive PBMC proliferation by 15% highlights MSCs' immunosuppressive potential. Upcoming immunophenotyping insights will detail the immunosuppression seen.
- Desenvolvimento e implementação de técnicas para deteção de alergénios nos alimentosPublication . Pequenas quantidades de um alergénio podem instigar uma cascata de reações imunológicas, mediadas por imunoglobulinas e, em casos graves, culminar na morte. Por esta razão, são considerados preponderantes para a segurança alimentar, dado que a frequência dessas reações tem aumentado a nível mundial. O processamento de alimentos pode aumentar o caráter alergénico dos alimentos, devido a alterações na estrutura e propriedades das proteínas, pelo que a rotulagem dos produtos depende de métodos sensíveis e específicos para a deteção de alergénios.
- Diagnostic pitfalls in Staphylococcus spp. surveillance: lessons from a multinational university student cohortPublication . Pinheiro, Bárbara; Gomes-Sampaio, Luís M.; Pacheco, Patrícia; Campos, Carla; Braz, Maria Pranto; Coelho, Cristina; Miranda, Carla; Quinteira, Sandra; Freitas, Ana R.; Campos, CarlaStaphylococcus aureus is a leading cause of severe and hard-to-treat human infections, particularly when resistant to cefoxitin due to the presence of the mecA gene [1]. However, diagnostic challenges arise from the misidentification of S. aureus and related species when relying on classical identification methods (mannitol fermentation;coagulase production), as well as from the detection of strains carrying the mecA gene but phenotypically susceptible to cefoxitin -known as "stealth" strains [2]. Building upon a previous collection of S. aureus from healthy students’ nares [3], we aimed to expand this collection with new samples, assess the occurrence of “stealth” isolates, and further investigate cases of incongruent identification. Nasal swab samples (n=557) from 507 students (median-23-years; 9 countries) attending alarge university (Porto district) were collected between March 2022 and November 2024. They were inoculated on to mannitol-salt agar and, in parallel, enriched in brain-heart-broth with 6.5%NaCl further plated on to ChromID® MRSA-SMART. Isolates deriving from mannitol-salt (only fermenting colonies) and chromogenic (all typical colonies) agar media were stored for species identification (MALDI-TOFMS), cefoxitin-susceptibility (disk-diffusion), and mecA gene screening (PCR). Staphylococcus aureus was identified in 46% (256/557; 6 countries) of cases. Other Staphylococcus species included S. haemolyticus(n=5), S. capitis(n=3), S. warneri(n=3), S. saprophyticus(n=1), S. simulans(n=1), and S. ureilyticus(n=1). These isolates expressed variable coagulase production (7 positive, 7 negative).On another hand, a non-fermenting S. aureus was detected (cromogenic medium).Ten (1.8%) students were colonized with methicillin-resistant staphylococci species carrying mecA including S. aureus (n=6), S. haemolyticus (n=3), S.ureilyticus (n=1). The mecA gene was also detected in 4/16 (25%; 3 Portuguese, 1 Italian) S. aureus susceptible to cefoxitin, the so-called “stealth”strains. Screening is ongoing in more isolates. Our study highlights the importance of integrating both phenotypic and genotypic methods for Staphylococcus accurate identification. Furthermore, the detection of stealth strains in healthy students underscores the need for robust community-based screening, as S. aureus carriage may be underestimated. Future studies will unveil whether these strains are capable of reversion to resistência.
