  • In vitro antibacterial and cytotoxic activity of Laurobasidium lauri extracts
Publication . Silva, L.; Oliveira, Ana Isabel; Moreira, Fernando; Ferraz Oliveira, Rita; Pinho, Cláudia
Plants and mushrooms have been used as medicines for many years, as a source of antibi-otics, antineoplastics, among others. Research have been conducted on the medicinal uses of Portu-guese plants, however, the therapeutic potentials of some of these plants used in traditional medicine, has remained unexploited. This study aims to evaluate antibacterial and cytotoxic activities, in vitro, of Laurobasidium lauri, a well-known fungus used in folk medicine on Madeira Island. An experimental study was performed using two extracts (aqueous and ethanolic 55% (V/V)) of the fun-gus isolated and in combination with three medicinal plants (Parietaria judaica, Polygonum aviculare, and Peperomia galioides). The antibacterial activity was evaluated against Escherichia coliand Staphy-lococcus aureus, through disc diffusion and broth microdilution methods. Cytotoxicity was evaluated using MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) assay. Regarding the disc diffusion method E. coliwas not susceptible to any of the extracts, except for the antibiotic (41.0 ± 1.0 mm). However,S. aureus, when subjected to 10 mg/ml of the ethanolic extracts of L. lauri(isolated and in combination) exhibited an inhibition with a diameter of 16,3 ± 2,5 mm and 12,0 ± 1,7 mm, respec-tively, when compared to the control, ciprofloxacin (24,0±1,0 mm). Also, the ethanolic extract of the isolated fungus had the best value of minimum inhibitory concentration (MIC) for S. aureus(MIC = 0.078125 mg/mL). The ethanolic extract of the fungus in combination with medicinal plants showed greater cytotoxic action on lung cancer cells A549 (IC50 = 48.3±1.0 μg/mL). The fungus presented cytotoxic and antibacterial potential, and the results observed may be related to some bioactive compounds (e.g., costunolide and dehydrocostuslactone, two natural sesquiterpene lactones present in Laurustrees, where the fungus grows). However, more research is needed to confirm these biological activities and mechanisms of action.

  • Phytochemical screening and antioxidant activity of green tea (Camellia sinensis) infusions commercialized in portugal
Publication . Silva, Inês; Gonçalves, Joana; Carmo, Juliana; Cruz, Agostinho; Oliveira, Ana Isabel; Ferraz Oliveira, Rita; Pinho, Cláudia
Natural antioxidant products, like green tea (Camellia sinensis) have gained popularity worldwide and new products claiming antioxidant activity are frequent in the dietary supplement industry. However, information regarding the antioxidant properties of these products, in the labels, is usually scarce. In addition, the antioxidant properties of commercial teas were not investigated in detail. Therefore, the present study aims to investigate the phytochemical and antioxidant activity of C. sinensis infusions.
  • Polypharmacy and drug interactions in older patients with cancer receiving chemotherapy: associated factors
Publication . Ferraz Oliveira, Rita; Oliveira, Ana Isabel; Cruz, Agostinho; Ribeiro, Oscar; Afreixo, Vera; Pimentel, Francisco
Polypharmacy in older adults with cancer receiving chemotherapy leads to increased risks of drug interactions, translating in potential hazardous health outcomes. This study aims to assess the prevalence of polypharmacy, drug–drug interactions (DDIs), and severe-drug interactions (SDIs) in older patients with cancer. Antineoplastic agents (ANAs) involvement and possible risk contexts (comorbidities with cardiac risk, and high-risk medications) were also analysed. ents (ANAs); it was conducted in three hospitals from the north of Portugal. Data collection was obtained using self-reports and medical records. DDIs were identifed and classifed using Micromedex® software. Descriptive and association analyze statistics were performed. Statistical hypothesis tests with p value less than 0.05 were considered signifcant. All statistical procedures and analysis were performed with R version 4.1.3. We enrolled 552 patients. Polypharmacy prevalence was 88.40%; 76.45% and 56.16% of the patients presented with DDIs and SDIs, respectively. SDIs with ANAs were found in 21.20% of the patients. High-risk medications were associated with a higher risk of polypharmacy, DDIs, and SDIs. Polypharmacy and DDIs were higher in patients with hypertension or diabetes. SDIs were higher in patients with diabetes. Polypharmacy, potential DDIs and SDIs were highly prevalent in older adults with cancer. A careful review of the medication administered is necessary to decrease it. These fndings warrant further research to optimize medication in this population and decrease problems related to medication, which may lead to emergency room visits and hospitalisations, compromising patient safety and/or ongoing treatments.
  • Physicochemical properties and antioxidant activity of portuguese craft beers and raw materials
Publication . Silva, Sara; Oliveira, Ana Isabel; Cruz, Agostinho; Ferraz Oliveira, Rita; Almeida, Rubim; Pinho, Cláudia
There is an increase in the popularity of craft beer, which is produced by small, independent, and traditional breweries. Since craft beer popularity is rising in Portugal this research focused on assessing physicochemical parameters, total phenolic content (TPC) and the antioxidant capacity of Portuguese craft beers and raw materials used in beer production. In this experimental study, 19 beer samples were analyzed. Parameters such as pH, Total Acidity, Reducing Sugar Content and TPC were evaluated. For the determination of antioxidant activity, DPPH scavenging activity and metal chelating activity (MCA) were analyzed in all samples. Craft beers demonstrated a high phenolic content (ranging from 343.78 mg GAE/L to 2172.49 mg GAE/L), significantly different from industrial beers. Craft beers demonstrated a higher inhibition of DPPH radicals and higher MCA than the raw materials. DPPH inhibition ranged from 36.5% to 96.0% for malt and 64.7% to 79.6% in hops samples. MCA also varied between the different samples, with results of 12.0% to 24.8% in malt samples and 3.8% to 23.5% in hops. Raw materials can potentially influence the antioxidant activity of the resulting beer. Positive correlations between TPC and physicochemical properties can be useful to help consumers choose beers with added value for health.

Publication . Santos, Roberta; Veiga, Inês; Cruz, Agostinho; Ferraz Oliveira, Rita; Pinho, Cláudia; Oliveira, Ana Isabel
