Fernandes, Filipe Jorge Ribeiro; Matos, Cristina Maria Fernandes Delerue Alvim deThe biggest obstacle to the treatment of diseases that affect the central nervous system (CNS) is the passage of drugs across the blood-brain barrier (BBB), a physical barrier that regulates the entry of substances into the brain and assures the CNS’s homeostasis. In this dissertation, a review was done on the lipid-based nanoparticles for the nanoencapsulation of neuroprotective compounds, thus increasing their bioavailability and capacity to cross the BBB. The gastrointestinal absorption was also discussed, as it may pose some obstacles into the absorption of the neuroprotective compounds into the bloodstream. The BBB’s composition was explained, as well as the transport mechanisms through which the compounds may cross into the brain parenchyma. A survey on the different types of lipid-based nanoparticles - nanoemulsions (NEs), nanoliposomes / nanophytosomes and solid lipid nanoparticles (SLNs) / nanostructured lipid carriers (NLCs) - was performed in this thesis, giving particular emphasis for their unique characteristics, the most important parameters influencing the formulation of each one, and some examples of studies that have been carried out in the encapsulation of neuroprotective compounds with the use of these nanoparticles.
